Single-Dose Psilocybin for a Treatment-Resistant Depression?

This article was published in the NEJM on Nov 3, 2022 where this study was done to assess the dose and safety of single-dose psilocybin formulation for a treatment-resistant major depressive episode. Psilocybin has shown efficacy in depression associated with cancer treatment, but can psilocybin help with treatment-resistant depression?

Dr. Harvinder Singh has summarized this recently published article in our Journal Club-19. Here is one slide from this jounral club discussion, summarizing the findings of this study:

RESULTS SUMMARY:

MADRS change from baseline to Week 3 (primary end point):

  • significantly better with a 25 mg dose than with a 1 mg dose
  • not significant difference between the 10 mg dose and the 1 mg dose

 

Incidences of response and remission at Week 3:

  • in the same direction as the primary end-point results

 

Sustained response at Week 12:

  • confidence interval for the odds ratio for 25 mg group as compared with the 1 mg group included 1
  • No definite conclusions can be drawn from secondary end-point results.
